When we question, tests, someone's belief, are we threatening something they like about themselves?

Often. It's why people often take it personally and why we have to take in to account the emotional dimensions of belief when we discuss them. However, other people to keep their beliefs at a emotional distance and respond very differently.

Depends on one's motive. I could question/test a belief because I'm looking to reveal its faults, or I could do so because I'm interested in adopting it.
